From surface coverage to biofilm involvement: Hyaluronic acid influences TiZr alloy corrosion under sterile and Staphylococcus aureus biofilm conditions.

Hyaluronic acid (HA) is widely used for intra-articular injection to improve joint lubrication and mobility, but its influence on microbiologically influenced corrosion of joint implants remains unclear. Here, we investigated how HA affects the corrosion behavior of TiZr alloy in simulated body fluid (SBF) under sterile and Staphylococcus aureus (S. aureus) biofilm conditions. Surface observations and chemical analysis indicated that HA-related surface coverage was formed on the TiZr surface and reduced surface damage under sterile conditions, suggesting a protective role of HA. However, in the presence of S. aureus biofilm, HA showed contrasting effects at different immersion stages, slightly reducing corrosion at the early stage compared with the S. aureus group but aggravating biofilm-associated localized damage after prolonged exposure. Contact angle analysis showed decreases of 68.1% and 57.1% within 1 min in the S. aureus + HA group on days 7 and 14, respectively, indicating that HA promoted rapid surface wetting and altered the interfacial wetting behavior in the biofilm environment. At these time points, electrochemical tests further confirmed corrosion acceleration, with the S. aureus + HA group showing the highest corrosion current densities among all groups, reaching 7.972 × 10-6 A cm-2 and 2.715 × 10-6 A cm-2, respectively. These results demonstrate that the effect of HA on TiZr alloy corrosion shifts from protection under sterile conditions to corrosion aggravation during prolonged S. aureus biofilm exposure. This finding suggests that HA-related effects on biofilm formation and implant corrosion should be considered when assessing the corrosion risk of TiZr joint implants under infection-related conditions.

Rhamnose-containing glycans and lipids differentially contribute to the Sporothrix-host interaction.

Rhamnose is a distinctive component of the cell wall of pathogenic Sporothrix species and plays important roles in host-pathogen interactions, including immune recognition, phagocytosis, and virulence. However, the mechanisms responsible for its incorporation into different surface glycoconjugates remain poorly understood. This study investigated the role of the putative rhamnosyltransferases RHT1 and RHT2 in Sporothrix schenckii and Sporothrix brasiliensis. RHT1- and RHT2-silenced mutants were generated in both species and analyzed for rhamnosyltransferase activity, cell wall composition, adhesion, biofilm formation, interactions with human immune cells, and virulence in the Galleria mellonella model. Cytokine production, TLR4-dependent recognition, and phagocytosis were evaluated to determine the contribution of distinct rhamnosylated glycoconjugates to host-pathogen interactions. Silencing of either gene reduced total rhamnosyltransferase activity but produced distinct effects on cell wall glycoconjugates. RHT2 silencing selectively decreased rhamnose incorporation into N-linked and O-linked glycans, whereas RHT1 silencing primarily altered the carbohydrate composition of a surface glycolipid, indicating functional specialization. RHT1-silenced mutants displayed reduced adhesion to HeLa cells and impaired biofilm formation in both species. Immune interaction assays revealed species-specific effects: in S. schenckii, RHT2 silencing had a stronger impact on cytokine production, TLR4-dependent recognition, and phagocytosis, whereas in S. brasiliensis, both RHT1 and RHT2 significantly contributed to these processes. Virulence assays showed that silencing either gene reduced virulence in S. schenckii, while virulence attenuation in S. brasiliensis was primarily associated with RHT1 silencing. RHT1 and RHT2 perform specialized, non-redundant functions in the biosynthesis of rhamnosylated glycoconjugates in Sporothrix. The distribution of rhamnose between glycoproteins and glycolipids differentially influences immune recognition, host colonization, and virulence, revealing species-specific contributions to the pathogenicity of S. schenckii and S. brasiliensis.

A Universal Quasi-Atom Interaction Model and Application in Designing Single-Atom Catalysts for Alkali-Metal Chalcogenide Batteries.

A deep understanding of reactant adsorption behaviors is crucial for unraveling structure-activity relationships and identifying simple descriptors for the kinetics of multi-electron redox reactions, such as the sulfur redox process in Li-S batteries (LSBs). Recently, the "10-electron rule" for evaluating the adsorption strength of single atoms (Eads) has been established for free-atom-like single-atom alloys (SAAs). However, our density-functional theory calculations show that this rule is unfeasible for single-atom catalysts (SACs) with metal centers covalently coordinated by nonmetal atoms. Herein, we proposed a new 18-electron rule for covalent SACs by establishing the quasi-atom interaction model (Q-AIM), which interprets the U-type behavior of Eads(S) dominated by antibonding-orbital filling. Additional calculations demonstrate that Eads(S) can serve as an efficient descriptor of the kinetics of sulfur redox involving multiatom polysulfides. Then, guided by the efficient predictor and Q-AIM, we employed machine learning to screen ∼800,000 candidate SACs. Ultimately, we obtained over 400 and 8000 promising SACs for sulfur evolution and sulfur reduction, respectively. Furthermore, Eads(S) can be extended to predict the catalytic performance of SACs in Li/Na/K-S/Se batteries. This work not only simplifies complex multi-electron processes to single-atom adsorption energy but also establishes a link between occupied electronic states and the catalytic performance of SACs.

Larger Owls (Aves: Strigidae) from the Lower Pleistocene of Crimea.

This paper describes large owls from the Lower Pleistocene of the Taurida Cave in Crimea, which are represented by three species. A series of postcranial skeletal elements is assigned to the tawny owl (Strix aluco)-this is the oldest find of this modern species. A fragmentary tarsometatarsus and femur represent a larger form-the fossil species Strix intermedia Jánossy, 1972, known mainly from the Middle Pleistocene of Western and Central Europe. The coexistence of S. aluco and S. intermedia within one fauna is shown for the first time and confirms the validity of S. intermedia. Furthermore, a new fossil species, Asio pythiotus sp. nov., is described based on a tarsometatarsus, morphologically similar to the long-eared owl (A. otus), but noticeably larger.

Advanced vs basic life support outcomes in treatment of out-of-hospital-cardiac-arrest: A registry-based cohort study.

The absolute effect on outcomes with Advanced, (ALS) as compared to Basic Life Support, (BLS) care in Out of Hospital Cardiac Arrest, (OHCA) remains unclear. To assess the association of ALS compared to BLS alone prehospital care on return of spontaneous circulation, (ROSC), survival to hospital discharge, and neurological outcome in OHCA in Detroit, MI. Retrospective analysis of 6746 patients from the Cardiac Arrest Registry to Enhance Survival database from January 2016 to December 2021 who had non-traumatic OHCA and received cardiopulmonary resuscitation. Cohorts were grouped by ALS v BLS care then initial rhythm and then compared by chi-squared, multivariable logistic, and multinomial regression analysis for outcomes of sustained ROSC, survival to hospital discharge and favorable neurological outcome at hospital discharge. The BLS cohort is defined by units that provided manual CPR, automated rhythm and defibrillation, airway management, but no endotracheal intubation or parenteral medications. ALS care was defined as manual rhythm interpretation, manual defibrillation, intravenous or intraosseous drug administration or endotracheal intubation. Mean age was 62.6 years (SD-16.7), with 56.3% male and 84.6% Black. Cohort breakdown: shockable ALS (S-ALS; n = 330, 4.9%); shockable BLS (S-BLS; n = 466, 6.9%); non-shockable ALS (NS-ALS; n = 2793, 41.4%); non-shockable BLS (NS-BLS; n = 3157, 46.8%). ROSC rates by cohort: S-ALS (37.9%); S-BLS (19.5%); NS-ALS (18.6%); NS-BLS (9.3%). ALS was associated with increased sustained ROSC for all initial rhythms (p < .001). S-ALS was associated with increased survival (p = .03), and neurologic outcome (p = .01) when compared to S-BLS. Advanced Life Support, relative to Basic Life Support alone, was associated with improved sustained return of spontaneous circulation across all initial rhythms, and improved survival to discharge and favorable neurologic outcome in shockable initial rhythms.

Psychometric properties of loaded and unloaded timed stair tests in assessing people with knee osteoarthritis.

To compare timed stair test performance between individuals with knee osteoarthritis and healthy older adults, examine reliability under loaded and unloaded conditions, explore correlations with functional measures, and identify optimal cut-off times. Cross-sectional study. Sixty individuals with knee osteoarthritis and 30 healthy older adults. Participants completed unloaded and loaded timed stair test trials 7 days apart. Knee strength, balance, mobility, and self-reported outcomes were assessed. Mean completion times for osteoarthritis participants were 23.91 s (unloaded) and 25.40 s (loaded). Intra-rater (ICC3,1 = 0.951-0.990 across raters and assessment days) and inter-rater (ICC3,2 = 0.999 [unloaded] to 1.000 [loaded]) reliabilities were excellent. Test-retest reliability was good for loaded (0.835) and excellent for unloaded (0.918) conditions. Times correlated negatively with affected side knee strength, balance, and confidence, and positively with pain severity, mobility tests, and daily activity difficulty. Optimal cut-offs were 18.67 s (unloaded) and 19.94 s (loaded), demonstrating sensitivities of 70.0% (unloaded) and 68.3% (loaded), a specificity of 86.7%, and area under the curve values of 0.78 (unloaded) and 0.77 (loaded). The timed stair test is a reliable, valid, and practical measure of functional performance in knee osteoarthritis, effectively distinguishing movement capability under loaded and unloaded conditions.
