Treatment of hyperlipoproteinemia, types II and IV, with pirifibrate: one hundred observations.

A new drug, pirifibrate, was investigated in a multicenter study of 100 patients with type IIa, IIb, or IV hyperlipoproteinemia (HLP). After one month of dieting, patients completed six months of treatment with a dose of 1,500 mg/day of pirifibrate. An average fall of about 20% in plasma cholesterol was observed in type IIa and IIb HLP, and more than 50% in the triglycerides in type IIb and IV HLP. Both reductions were statistically significant (P less than 0.00005). Increases in the alpha-lipoproteins were observed in the three types of HLP. The pre-beta-lipoproteins showed a statistically significant fall in type IIb and IV HLP. Variations observed in beta-lipoproteins were significant only in some controls with type IV, HLP, who had low basal levels.

Pirifibrate in the treatment of hyperlipoproteinemia: types IIa, IIb, and IV.

Pirifibrate was given to 53 patients with type IIa, IIb or IV hyperlipoproteinemia (HLP), and the results of the first three months of treatment were evaluated. Patients in the study had cholesterol and triglyceride plasma levels higher than 300 mg/100 ml and 200 mg/100 ml, respectively, after following a diet without any pharmacological treatment for one month. After treatment with pirifibrate, mean plasma levels of cholesterol fell about 20% in types IIa and IIb HLP; triglycerides fell between 30% and 46% in types IIb and IV HLP. Slight increases in alpha-lipoproteins in the three types of HLP were measured. Prebeta-lipoprotein levels fell considerably in types IIb and IV. The variations observed in beta-lipoprotein levels were significant only in type IV, which initially showed low values.

Pirifibrate in the treatment of hyperlipoproteinemias: a multicenter study.

Pirifibrate, a new hypolipemic substance, was studied in a multicenter trial for three months in 151 patients diagnosed as having type IIa, IIb or IV hyperlipoproteinemia (HLP). The patients admitted into the study had not received any hypolipemic medication for four weeks immediately before the study. We observed falls of between 24.3% and 20.0% in the average levels of plasma cholesterol in types IIa and IIb HLP. The triglycerides showed a decrease in average values, which varied between 31.0% and 38.6% in types IIb and IV HLP. The alpha-lipoproteins always increased in three types of HLP studied, while the pre-beta-lipoproteins showed a fall in averages in types IIb and IV HLP. The beta-lipoprotein levels were reduced in type IIa HLP and reached statistical significance.
