Biodistribution of Phosphorothioate-Based Antisense Oligonucleotides in Stabilin Receptor Knock-Out Models.

Phosphorothioate-based antisense oligonucleotides (PS-ASO) are a common class of ASO for therapeutic uses due to their resistance to nucleases and known deposition in human physiological systems. On the molecular level, these ASOs are recognized by several scavenger receptors, including scavenger receptors A and B (SR-A, SR-B) and Stabilin receptors, Stab1 and Stab2. The Stabilins bind and internalize a host of endogenous and exogenous molecules with high endocytic rates and low turnover times. To determine their specific role in the biodistribution of PS-ASOs, we intravascularly injected knockout (KO) mice for Stab1, Stab2 or Stab1Stab2 double KO (DKO). Our results show that each receptor contributes to PS-ASO clearance from the blood and accumulation in liver and spleen, locations where these receptors have the highest levels of expression. Moreover, Stab2 is the primary clearance receptor taking up the bulk of the PS-ASO found in blood. Our conclusion is that Stabilins have a profound effect on the catabolism of PS-ASOs, for which internalization and clearance from the tissue need to be taken into consideration in the design of any therapeutic ASO.

Domain Knowledge Constrained Symbolic Regression for Optimising Dermal Drug Formulations.

Interpretable dynamic models that remain physically admissible under extrapolation are essential for dermal formulation optimisation. Mechanistic models are often labour-intensive to derive, whereas unconstrained data-driven models may violate physical constraints and degrade beyond observed conditions. We developed a domain knowledge constrained symbolic regression (DKC-SR) framework to discover compact, interpretable dynamic models for cumulative release directly from time-series experiments. Domain knowledge was embedded through a restricted operator set and feasibility constraints over the bounded optimisation domain. Candidate expressions were evaluated by numerically solving the dynamic differential equations and selected using an information-criterion trade-off across a level-wise set of complexity-controlled models. The final surrogate was coupled with the covariance matrix adaptation evolution strategy for formulation optimisation. In an ibuprofen formulation case study, the selected model provided a formulation ranking that, combined with experimental validation, guided the optimiser to a narrow high-performing region; a representative optimum was experimentally confirmed to exceed the historical best. Although the surrogate was more reliable for ranking and optimisation guidance than for exact quantitative calibration, it remained physically admissible and numerically stable across the design space. DKC-SR provides a compact, interpretable, and optimisation-ready route to modelling dermal release under bounded design constraints.

Novel zirconium-based supramolecular organic frameworks/fullerene@strontium-based MOFs composite for highly sensitive electrochemical sensing of ciprofloxacin.

A novel ternary composite of zirconium-based supramolecular organic framework/fullerene@strontium-based metal-organic framework (Zr-SOF/C60@Sr-MOF) was synthesized and adopted as an electrode modifier to construct a high-sensitivity electrochemical sensor for ciprofloxacin (CIP) determination. The composition and morphology of the material were confirmed by SEM, TEM, XPS, XRD and FT-IR. Cyclic Voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were used to evaluate the electron-transfer properties and interfacial behavior of the modified electrodes, and square wave voltammetry (SWV) was employed for the sensitive and quantitative detection of the target analyte. The results reveal that the composite inherits the abundant porous structure of Zr-SOF, the expansive specific surface area of Sr-MOF and excellent electron transfer capability of fullerene. The sensor enables accurate CIP detection in the linear range 5.0 nM-1000 µM, with a detection limit of 7.0 nM (S/N = 3), along with satisfactory selectivity, reproducibility and long-term stability. Practical application was validated in lake water, tap water and milk samples, yielding recoveries of 95.3%-101.63%. This work provides new insights into SOF/MOF-based electrochemical sensing and establishes an efficient strategy for rapid on-site detection of CIP antibiotics in environmental and food matrices.

Induction and Propagation of Ferroptosis Evasion by FTH1P7 Drives Sorafenib Resistance in Hepatocellular Carcinoma.

Sorafenib refractoriness severely compromises therapeutic efficacy in hepatocellular carcinoma (HCC). This study identifies ferritin heavy chain 1 pseudogene 7 (FTH1P7) as a critical driver and propagator of sorafenib resistance. FTH1P7 was significantly upregulated in HCC cells resistant to sorafenib. Mechanistically, the self-sustaining FTH1P7/microRNA-182-5p/FOXC1 positive feedback loop maintained high FTH1P7 expression levels in resistant cells. Elevated FTH1P7 functioned as a competing endogenous RNA to the mRNA of its parental gene, ferritin heavy chain 1 (FTH1). Increased FTH1 levels enhanced Fe2+ oxidation, thereby inhibiting ferroptosis-a key anticancer mechanism of sorafenib-thereby conferring resistance. Furthermore, exosomes derived from resistant cells transferred FTH1P7 to sorafenib-sensitive cells, thereby horizontally propagating resistance. To counter sorafenib resistance, we developed a nanoliposome co-delivering sorafenib and FTH1P7-targeting siRNA (SR-siFTH1P7-LIP). This nanomedicine synergistically and effectively silenced FTH1P7 expression and reversed resistance. These results elucidate novel mechanisms of sorafenib resistance, involving FTH1P7-mediated induction and exosomal propagation of ferroptotic resistance, and propose a promising nanomedicine-based strategy to combat sorafenib refractoriness in HCC.

Methodological Rigor and Integrity of Systematic Reviews and Meta-Analyses From Nursing-Affiliated Institutions in North African Countries: The FALCON-1 Study.

Systematic reviews (SRs) and meta-analyses (MAs) are central to evidence-based practice (EBP). Despite their growing volume in low- and middle-income countries, especially those produced by nursing-affiliated institutions in North Africa (NA), no empirical meta-research has systematically assessed their methodological quality, reporting practices, and research integrity indicators. This study investigated methodological quality, reporting practices, and scientific integrity of SRs and MAs conducted in this context over the past decade. We conducted a meta-research synthesis of SRs and MAs in nursing sciences published between 2015 and 2025 using the PubMed database. Eligible reviews used explicit SR/MA methods, had a first or last author affiliated with a NA institution, and were identified through structured searches. Methodological and reporting quality were assessed using PRISMA and MOOSE guidelines, complemented by an exploratory 10-point composite quality score. Associations between review characteristics and quality indicators were examined. We included 38 SRs/MAs, of which 15 included quantitative synthesis. Only 50% reported PROSPERO registration, with several registrations being retrospective or inaccurate. While eligibility criteria, database searches, and quality appraisal were commonly reported, dual data extraction and gray-literature searches were less common. Validated appraisal tools were inconsistently applied, and reporting guidelines were sometimes inappropriately used. The composite score indicated overall modest methodological quality; with PROSPERO registration was the only factor associated with higher quality scores (p = 0.008). Heterogeneity was usually reported, whereas sensitivity analyses and publication-bias assessments were inconsistently conducted. This study highlights important methodological and integrity-related deficiencies in SRs and MAs in NA. Strengthening methodological expertise in this central field of clinical practice is critical.

[Saposhnikoviae Radix enhances resistance to respiratory tract infection by potentiating interferon signaling].

This study aimed to investigate the immunopharmacological mechanism of Saposhnikoviae Radix(SR) against respiratory viruses and bacteria infections based on immune signal activation. Mouse models of pneumonia induced by intranasal influenza A virus(H1N1) infection and intratracheal lipopolysaccharide(LPS) instillation were established. The effects of the water extract of SR on pulmonary CT imaging, histological damage, and inflammatory cytokine expression in lung tissues were evaluated. Using bone marrow-derived macrophages, this study analyzed the regulatory effects of the extract on macrophage proliferation and phagocytic function. RNA-seq and bioinformatics analysis, combined with molecular biology experiments, were employed to explore the regulatory role of the water extract of SR on the type Ⅰ interferon(IFN-Ⅰ) signaling pathway. The findings revealed that the water extract of SR significantly alleviated H1N1 infection-induced lung imaging and pathological damage in mice, reduced lung viral gene copies, and downregulated the expression of inflammatory cytokines such as interleukin-6(IL-6), tumor necrosis factor-α(TNF-α), and CXC motif chemokine ligand 10(CXCL10). Concurrently, drug treatment demonstrated similar lung tissue protective functions in the LPS-induced mouse pneumonia model. Cell experiments showed that the extract promoted proliferation of macrophages and enhanced their phagocytic capacity toward neutral red. Transcriptomic analysis revealed that genes upregulated by the extract of SR were significantly enriched in pathways including H1N1 infection, Toll-like receptor signaling, NOD-like receptor signaling, and IFN signaling. Finally, qRT-PCR and Western blot confirmed that the extract activated the interferon signaling pathway. In conclusion, the extract of SR activates innate immunity centered on IFN-Ⅰ signaling and enhances macrophage function, thereby improving the host's defense against viral and bacterial infections. It possesses potential preventive and therapeutic value for respiratory tract infection-associated pneumonia and lung injury.
