Morphological and Multilocus Phylogenetic Characterization of Septoria Species from Dioscorea, Eclipta, and Taraxacum in East Asia.

More than 90 species of Septoria have been reported from Korea, but the identity of many recorded taxa remains uncertain owing to the lack of detailed morphological documentation and multilocus sequence data. In this study, isolates collected from symptomatic leaves of economically and medicinally important plants between 2006 and 2024 were examined using morphological observations and multilocus phylogenetic analyses based on six loci (internal transcribed spacer (ITS) region, actin (ACT), β-tubulin (Btub), calmodulin (CAL), elongation factor 1-alpha (EF1-α), and RNA polymerase II second largest subunit (RPB2)). The concatenated dataset resolved three well-supported lineages corresponding to Septoria dioscoreicola on Dioscorea polystachya, S. eclipticola on Eclipta prostrata, and S. miurae on Taraxacum officinale, of which S. dioscoreicola and S. eclipticola are newly recorded in Korea, whereas Korean materials previously reported as S. taraxaci on Taraxacum are re-identified here as S. miurae. Multilocus analyses further demonstrated that S. taraxaci and S. miurae represent two distinct species associated with Taraxacum, with Korean and Japanese isolates corresponding to S. miurae, whereas the third Taraxacum-associated taxon, S. taraxacicola, is regarded as a synonym of S. miurae. Given the economic and medicinal importance of these hosts involved, the occurrence of Septoria leaf spot on these species may represent a potential concern for their cultivation in East Asia. These results provide an updated taxonomic basis for Septoria species occurring in Korea and clarify species boundaries within the Taraxacum-associated complex.

Two new species of the genus Sinocrassula (Crassulaceae) from Yunnan Province, China.

Based on comprehensive morphological and molecular data analyses, this study confirms that three populations of Sinocrassula from central Yunnan Province represent two distinct new species, which are described herein as Sinocrassula kunmingensis J.Guan Wang & Jing Zhao and S. chenggongensis J.Guan Wang & Jing Zhao. Morphologically, the two new species are most similar to S. adpressa, S. crassifolia, S. obliquifolia, and S. yongshengensis. However, S. kunmingensis is characterized by its greenish-yellow or red stem leaves without spots, subligulate nectar scales, reddish-purple flowers, ovate-lanceolate bracts, and petals with a slight central protrusion; S. chenggongensis is distinguished by its green stem leaves with purplish-red spots, subligulate nectar scales, pinkish-purple flowers, oblong-elliptic bracts with an oblique apex, and petals without a protrusion. This combination of morphological characters can be clearly distinguished from those of similar species. Phylogenetic analyses further reveal that the two new species form two independent clades with strong support and are sister to each other.

Lacticaseibacillus rhamnosus GG lysate inhibits Staphylococcus aureus biofilm formation in human skin explants.

Staphylococcus aureus is a leading cause of skin and soft tissue infections and a major nosocomial pathogen, with biofilm formation contributing to significantly higher morbidity and mortality. In atopic dermatitis (AD), where skin barrier dysfunction and microbial dysbiosis are common features, S. aureus abundance and biofilm formation are associated with disease severity and flare recurrence. We previously demonstrated that a lysate of Lacticaseibacillus rhamnosus GG (LGG)- a common human commensal and widely used probiotic strain- can inhibit S. aureus attachment to human keratinocytes by mechanisms including displacement and competitive exclusion. In this study, we extended these findings utilizing organ-cultured human skin to evaluate the protective effects of LGG lysate against S. aureus challenge in a physiologically relevant model. LGG lysate preserved skin integrity by preventing S. aureus penetration, proliferation, eDNA release and biofilm maturation. These effects were dose-dependent and effective when the lysate was applied from 24 hours pre-S. aureus inoculation to up to 3 hours post inoculation. Together, these data provide mechanistic insight into microbiome- pathogen interactions at the skin surface which could be exploited for therapeutic potential, particularly in the prevention of nosocomial S. aureus skin infections and mitigating S. aureus-driven flares in AD.

Subcutaneous Implantable Cardioverter-Defibrillator Therapy in Women.

The subcutaneous implantable cardioverter-defibrillator (S-ICD) is an established therapy for sudden cardiac death prevention, but sex-specific outcomes remain incompletely characterized. This study evaluated sex-related differences in baseline characteristics, appropriate shocks, complications, reinterventions, and mortality among S-ICD recipients. The nationwide HONEST (S-ICD French Cohort Study) cohort enrolled all patients who received an S-ICD in France between 2012 and 2019. Clinical endpoints were centrally adjudicated. Sex-specific associations with outcomes were assessed by using propensity score-based inverse probability weighting. Among 4,924 S-ICD recipients, 1,148 were women (23.3%). Compared with men, women were younger (47.3 ± 15.6 years vs 50.6 ± 14.7 years; P < 0.001), less frequently received an implant for primary prevention (57.6% vs 65.1%; P < 0.001), less often had coronary artery disease (38.9% vs 56.5%; P < 0.001), and more often had electrical heart disease (26.3% vs 20.4%; P < 0.001). After adjustment, women had a lower 5-year risk of appropriate shocks (HR: 0.85; 95% CI: 0.74-0.98; P = 0.023) and similar overall complication and reintervention rates but a distinct complication profile, with higher risks of chronic pain (HR: 2.63; 95% CI: 1.61-4.29; P < 0.001) and lead dislodgment (HR: 1.79; 95% CI: 1.09-2.95; P = 0.022) and a lower risk of inappropriate shocks (HR: 0.64; 95% CI: 0.50-0.82; P < 0.001). All-cause mortality was lower in women, whereas S-ICD-unresponsive sudden death and device-related mortality were similar. Women receiving an S-ICD experienced fewer appropriate shocks, with similar overall complication and reintervention rates, but a distinct complication profile. These findings support sex-informed S-ICD selection and follow-up. (S-ICD French Cohort Study [HONEST]; NCT05302115).

The conserved QTQTX motif in the SARS-CoV-2 spike protein is dispensable for cleavage and lung cell entry of the emerging variant BA.3.2.

The furin motif in the SARS-CoV-2 spike (S) protein is important for lung cell entry. It is embedded in an extended loop structure and preceded by a highly conserved QTQTX motif that is required for efficient furin cleavage of the SARS-CoV-2 WA-1 S protein. BA.3.2 is an emerging SARS-CoV-2 saltation variant that is spreading globally in April 2026 and encodes a highly mutated S protein. Here, we analyzed whether the QTQTX motif is also required for spike protein cleavage and lung cell entry of BA.3.2. We report that two patient-derived spike sequences of the BA.3.2 subvariant BA.3.2.2 lack the first QT repeat of the QTQTX motif and show that this motif is largely dispensable for both cleavage and lung cell entry of BA.3.2.2, which we found to depend on TMPRSS2. Our results suggest that the reconfiguration of the BA.3.2 S protein during persistent infection may have significantly altered the determinants of furin cleavage.IMPORTANCEThe furin motif in the SARS-CoV-2 spike (S) protein is unique among sarbecoviruses and constitutes a virulence determinant. A QTQTX motif located immediately upstream of the furin motif is required for furin cleavage of the S protein of the virus that circulated early in the pandemic. Here, we show that the QTQTX motif is largely dispensable for S protein processing and S protein-driven lung cell entry of the emerging saltation variant BA.3.2, which is currently spreading globally. Thus, BA.3.2 evolution within immunocompromised individuals may have relaxed the requirements for furin processing of the spike protein.

A case of severe strongyloidiasis complicating immunosuppressive therapy initiated for IgA vasculitis.

IgA vasculitis (IgAV) and Strongyloides stercoralis infection may both present with abdominal pain and purpura; however, glucocorticoid, the treatment for IgAV, is a major risk factor for S. stercoralis hyperinfection syndrome and disseminated strongyloidiasis. To date, only 2 cases of IgAV with concurrent S. stercoralis infection have been reported globally, both in children. We report a 72-year-old man presenting with abdominal pain and purpura. Diagnosed with IgAV during this admission, he received glucocorticoid therapy. However, the patient's condition did not improve as expected. Subsequently, S. stercoralis was detected in the patient's sputum and stool. We report the first adult case of IgAV with concurrent S. stercoralis infection, highlighting the importance of screening patients with IgAV for strongyloidiasis before initiating immunosuppressive therapy.
